Abstract:
[PROBLEMS] To provide a method for preparing a heavy atom derivative of a protein crystal in a simple manner with high probability, a method for collecting phase information, and a method for determining a three-dimensional structure of a protein crystal. [MEANS FOR SOLVING PROBLEMS] The method for preparing a heavy atom derivative of a protein crystal comprises allowing iodine to act on a protein crystal. Vaporized iodine and hydrogen peroxide are allowed to act on the protein crystal to prepare a heavy atom derivative. The method for collecting phase information comprises subjecting the heavy atom derivative of the protein crystal prepared using iodine by the above method to a X-ray diffraction test to collect phase information of diffraction X-ray from the heavy atom derivative. The method for determining a three-dimensional structure of a protein crystal comprises determining the three-dimensional structure of a protein crystal based on the phase information of diffraction X-ray from the heavy atom derivative collected by this method.
