Abstract:
The present invention addresses the problem of developing a technique for producing a novel modified protein ligand that maximizes binding capacity and binding efficiency with respect to a target molecule when said ligand is immobilized via a lysine residual (side-chain epsilon-amino group) that allows an efficient carrier-immobilization reaction. Provided are: a modified protein containing a sequence comprising protein A, the typical protein ligand used for affinity purification, mutated by substitution of all of the lysine residuals therein with other amino acids and with lysine attached to the end thereof; and an affinity-separation matrix wherein said modified protein is immobilized on a water-insoluble carrier via reductive amination or the like. This affinity-separation matrix is characterized by high binding capacity with respect to a target molecule even with a low quantity of immobilized ligands.
