Abstract:
A novel proteome analysis method whereby quantitative changes of proteins expressed in cells are monitored with the passage of time and thus the genomic expression state can be quantitatively analyzed. This method comprises adding a liquid culture medium containing an isotope-labeled amino aid to vital cells, culturing the cells and then collecting the same after a definite period of time, extracting proteins therefrom, fractionating the extracted proteins by developing by two-dimensional electrophoresis, and quantifying arbitrary proteins thus fractionated each based on the mass difference betweem a protein having the labeled amino acid and the protein free from the labeled amino acid.
