Abstract:
Provided in the present invention is a rapid next-generation sequencing method, which comprises the following steps: polymerization, involving: a detection complex is linked to a primer hybridized to a nucleic acid molecule to be detected, wherein the detection complex is modified with a fluorescent group and a blocking group; acquisition, involving: the fluorescent group is excited to generate a fluorescence signal, an imaging spot is obtained by means of an optical imaging system, and the imaging spot is identified to obtain the type of a base; excision, involving: the blocking group and the fluorescent group are excised; and the repeating of the polymerization-acquisition-excision cycle, wherein the acquisition is started after the step of polymerization is started, and/or the excision is started after the step of acquisition is started. In the sequencing method of the present invention, unfinished synthesis reactions and the preparatory work of excision reactions are performed during the signal acquisition period that needs to be consumed for a long period of time, which compresses the time of a single cycle, thereby finally shortening the time of the whole sequencing process.
