Abstract:
Disclosed are recombinant Escherichia coli for efficient production of succinic acid and a construction method for the recombinant Escherichia coli. The method comprises: knocking out a by-product encoding gene of a succinic acid producing strain E.coli FMME-N-5 to obtain a strain E.coli FMME-N-5(ΔfocA-pflB-ΔldhA-Δpta-ackA); and introducing a plasmid pTrcHisA-pck-ptxD into the strain, overexpressing actinobacillus succinogenes phosphoenolpyruvate carboxykinase pck and bacillus stutzeri phosphite dehydrogenase ptxD, and screening to obtain an engineering bacterium E.coli FMME-N-5(ΔfocA-pflB-ΔldhA-Δpta-ackA)-pck-ptxD for efficient production of succinic acid, the engineering strain using a two-stage fermentation strategy on a 7.5 L fermentation tank for fermentation for 96 h, the succinic acid yield reaching 137 g/L, the yield of succinic acid reaching 1 g/g glucose, the production strength being 1.43 g/L/h, by-products lactic acid and formic acid being not accumulated, and acetic acid being 1-2 g/L.
