Abstract:
Disclosed are a recombinant plasmid combination, a genetically modified yeast strain, and a method for producing an odd chain fatty acid. The recombinant plasmid combination comprises at least one expression plasmid, the expression plasmid comprising at least one polynucleotide encoding a polypeptide having a threonine deaminase activity, a fatty acid synthase activity, a propionyl-CoA synthase activity, an aldehyde dehydrogenase activity, or a keto acid decarboxylase activity. The recombinant plasmid combination can be used to genetically modify a microorganism, such that the expression of key enzymes can be regulated so as to establish a synthesis route for synthesizing an odd-chain fatty acid in the modified microorganism. Propionic acid, or L-threonine and/or glucose can be used to directly synthesize the odd-chain fatty acid, thus keeping fermentation costs low. The yield and proportion of the odd chain fatty acid can be further increased by continual application of the genetic modification process.
