Abstract:
Disclosed is a screening method which can screen for a prophylactic and/or therapeutic agent for a disease associated with a hepatitis C virus efficiently in a simple manner. Also disclosed is a prophylactic and/or therapeutic agent for a disease associated with a hepatitis C virus, which is produced by the method. The screening method for a prophylactic and/or therapeutic agent for a disease associated with a hepatitis C virus comprises the step of evaluating the inhibitory activity of a substance of interest on the interaction between a hepatitis C virus core protein and PA28γ. The method may be conducted by employing the transcriptional activity of a lipid metabolism regulation factor as a measure. The substance to be tested in the method may have an inhibitory activity on the expression or function of PA28γ gene. The disease associated with a hepatitis C virus includes hepatitis C, fatty liver, acute hepatitis, chronic hepatitis, cirrhosis, liver cancer and an insulin-resistance disease. The method is extremely useful as a method for producing an agent having an excellent prophylactic and/or therapeutic effect particularly on fatty liver or liver cancer with efficiency.
