Abstract:
Disclosed in the present invention is a sequence image super-resolution correction method based on image gradient variance analysis. The present invention is used for processing a fluorescence sequence image captured by an optical microscope, the grayscale value distribution of the image reflects the distribution of the fluorescence signal intensity, and a time sequence fluctuation feature is recorded as time changes. The method in the present invention comprises: firstly, analyzing two types of information of a sequence image: one involving, for fluorescence intensity distribution, analyzing a variance of the fluorescence intensity of each pixel point itself as time changes, and the other involving calculating a fluorescence intensity gradient field and analyzing a variance of the fluorescence intensity gradient of each pixel point as time changes; then defining a weighting function by using the two variances, and performing weighted correction on an original sequence image; and finally, performing SRRF analysis on the corrected image sequence to obtain a super-resolution sample fluorescence image having low background noise, a weakened artifact and an enhanced resolution capability. The method has a wide applicability, and can be applied to a common wide field, confocal imaging, structured light illumination imaging, etc.
