Abstract:
A site-occupying primer used in sequencing and a removal method. The site-occupying primer comprises a blocking group at the 3' end. The 3' end sequence of the site-occupying primer is complementary to a part of the sequence of a sequencing template, and the 5' end sequence of the site-occupying primer is not paired with the sequencing template. A method for removing a primer and a related sequencing method. A section of sequence not paired with the template is introduced at the 5' end of the site-occupying primer, and the branch migration action of DNA is used to easily remove the site-occupying primer when the site-occupying
primer is required to be removed.
