Abstract:
This sugar chain analysis method for analyzing a sugar chain structure bound to an antibody, involves: a release step in which a sugar-cleaving enzyme is added to a sample that contains an antibody, and the sugar chains bonded to the antibody are specifically released to obtain free sugar chains; a recovery step in which the free sugar chains are recovered by affinity purification; a hydrophobization step in which the free sugar chains are hydrophobized to obtain hydrophobized sugar chains; and a measurement step in which the hydrophobized sugar chains are subjected to supercritical liquid chromatography and tandem mass spectrometry, in that order.
