Abstract:
A super-resolution microscopic imaging method and apparatus based on a shared-path parallel fluorescence emission difference. Exciting light during fluorescence emission difference microscopy imaging is modulated by using a liquid crystal spatial light modulator (6). Two parts of the liquid crystal spatial light modulator (6) are respectively loaded as 0-2 π vortex phase modulation and a blazed grating, such that a solid light spot and a hollow light spot, which are at a certain distance from each other, are simultaneously formed on a sample surface by exciting light that shares a path, and are subjected to parallel scanning, thereby doubling the imaging speed compared with common fluorescence emission difference microscopy imaging, and also making two beams of exciting light not be susceptible to noise and drift interference due to sharing of the path.
