Abstract:
The purpose of the present invention is to provide: a support onto which Protein A has been immobilized, the Protein A having a specific amino acid sequence that enables the dissociation of antibodies under mild pH conditions (specifically, pH4.0-5.5) in which many antibodies do not undergo acid modification; and a manufacturing method for the support. An immobilization support (excluding an immobilization support having a monolith structure) onto which a protein is adsorbed by electrostatic interaction, said protein comprising an amino acid sequence represented by general formula R1-R2-R3-R4-R5-R6, and the section of said protein represented by R1-R2-R3 being used for immobilization on the immobilization support (in the formula: the sequence represents a sequence from the amino-terminal side towards the carboxyl-terminal side; the sequence of the R2 part is either a protein A mutant sequence, which is the protein to be immobilized, or a sequence in which 1-3 of said sequences are linked; and the protein A mutant exhibits the characteristics of strongly binding to the antibodies in neutral conditions, and dissociating with the antibodies which were bonded with in the neutral conditions when in mildly acidic conditions in the range of pH4.0-5.5].
