Abstract:
A purification system and method used for continuous purification. The purification system comprises: (1) at least one affinity chromatography main unit, wherein each affinity chromatography main unit independently comprises one or more affinity chromatography sub units; (2) at least one virus inactivation main unit, wherein the virus inactivation main unit independently comprises one or more virus inactivation sub units; and (3) at least one concentration and/or liquid replacement main unit, wherein each concentration and/or liquid replacement main unit independently comprises one or more concentration and/or liquid replacement sub units. The upstream or downstream of each main unit independently and optionally has one or more buffer volumes; each main unit and each buffer volume are connected by means of a pipeline through which a feed liquid may circulate; at least one weight sensor and/or at least one liquid level sensor are/is configured for each buffer volume; the weight sensor and the liquid level sensor are in communication connection with a management system; the management system controls the startup, continuation, and stop of the purification system according to the signal of the weight sensor and/or the liquid level sensor; a target molecule purified by the purification system is a protein; a concentration and/or liquid replacement unit is used for performing continuous operations to perform ultrafiltration or dialysis; and in the concentration and/or liquid replacement unit, the flow direction of a stream comprising a sample is different from a filtration direction.
