Abstract:
Provided is a system for producing an AAV genetic medicine by infection of an insect cell using a baculovirus. The system comprises: a first baculovirus, a second baculovirus, a third baculovirus, and an Sf9 cell; an expression cassette encoding an Rep78 protein and an expression cassette encoding a Cap protein VP2 are integrated on the genome of the first baculovirus, wherein the expression of the gene encoding the Rep78 protein is driven by a polyhedrin promoter, and the expression of the gene encoding the Cap protein VP2 is driven by a p10 promoter; an expression cassette encoding an Rep52 protein and an expression cassette encoding a Cap protein VP1 are integrated on the genome of the second baculovirus, wherein the expression of the gene encoding the Rep52 protein is driven by the polyhedrin promoter, and the expression of the gene encoding the Cap protein VP1 is driven by the p10 promoter; an AAV vector sequence carrying a target gene expression cassette is integrated on the genome of the third baculovirus; and an AAV carrying a target gene is produced by co-infection of the Sf9 cell using the first baculovirus, the second baculovirus, and the third baculovirus.
