Abstract:
Provided is a novel target gene capture method (CPCS) based on CRISPR technology, which comprises: using a sgRNA Cas protein complex to cut and capture a target gene sequence, and meanwhile, digesting a non-target gene sequence with nucleic acid hydrolase to obtain a target gene sample. According to the method, the CRISPR technology is used in combination with the nucleic acid hydrolase for target gene capture, the target gene sequence is separated from the non-target gene sequence by utilizing the characteristics of precise positioning and cutting of CRISP, and meanwhile, the nucleic acid hydrolase is used for hydrolyzing the non-target gene to obtain the target gene by utilizing the characteristics that the Cas protein remained at the cutting site after cleavage. The method has the characteristics of low cost, high specificity, and short-time capture, and by the method, it takes as little as 5 hours to complete the processes of target gene capture and library construction.
