Abstract:
A tiling light sheet selective planar illumination microscope (TLS-SPIM), a use method thereof, and a microscope system. The use method comprises: step (101), loading a corresponding phase map (201, 202) to each group of pupil subsections (205, 206) of the pupil by means of a spatial light modulator (401), and performing phase modulation on an excitation beam to create at least two coaxial excitation beam arrays (203, 204); step (102), scanning the created at least two coaxial excitation beam arrays (203, 204) to generate discontinuous light sheets (1002) accordingly; and step (103), tiling at least one of the generated discontinuous light sheets (1002) in the propagation direction of the excitation light to obtain tiling light sheets for selective planar illumination of a sample. The utilization of the use method of the TLS-SPIM, the TLS-SPIM and a system comprising same enables the increasing of the imaging speed, the improvement of the resolution, and the reduction of the source data amount.
