Abstract:
The present invention provides a UdgX-based technology for detecting DNA deoxyuracil on a single-base resolution level. Specifically, the present invention provides a UdgX cross-linking and polymerase stalling sequencing ("Ucaps-seq") technology, thus achieving the detection of dU in single-base-resolution DNA. The Ucaps-seq method of the present invention is a powerful tool, and has the advantages of high sensitivity, high specificity, high resolution, etc., and has broad potential application, particularly in aspects such as disease diagnosis, evaluating gene editing accuracy, etc.
