An ultrasonic microbubble cell stimulation device and an operation method thereof. The ultrasonic microbubble cell stimulation device comprises: a culture plate (11), a positioning frame (12), and a base (13). The culture plate (11) is internally provided with a culture well (14) and is provided with a sealing membrane (10) at an open end thereof. The culture well (14) is internally provided with a culture cup (15) that is for injecting microbubbles into the culture well (14) and is capable of preventing the mixture of air and the microbubbles when the culture plate (11) is sealed by the sealing membrane (10) and inverted. The base (13) is connected with a transmission component (16) that is for transmitting an ultrasonic wave to stimulate a cell and is capable of adjusting a distance from the base to the culture plate (11). The base (13) and the positioning frame (12) are connected through a position-adjusting component (17). The operation method of the ultrasonic microbubble cell stimulation device comprises: a guide rod (171) installation step; a transmission component (16) connecting and assembly step; a positioning frame (12) installation step; a culture cup (15) installation and microbubble injecting step; and a culture plate (11) installation step. The ultrasonic microbubble cell stimulation device can successfully prevent air bubbles from entering into the culture plate (11), ensuring that the ultrasonic waves act on the microbubbles directly, has a controllable stimulation area and intensity, is easy to operate, is versatile, and has a wide range of application.
