Abstract:
Disclosed are the use of the MATE1 gene and/or protein as a treatment target of colorectal cancer and the use thereof for determining a marker for a treatment plan of colorectal cancer. Also disclosed is the use of the MATE1 protein and/or inhibitor of the encoding gene SLC47A1 thereof in the treatment of colorectal cancer. The down regulation of the expression of MATE1 in the discharge passage of metformin is a key mechanism for the sensitivity of KRAS mutant colorectal cancer cells to metformin; and the transcriptional level of MATE1 is decreased and the concentration of metformin in tumor cells is increased by upregulating the methyltransferase DNMT1 and down-regulating the demethylase TET1/2, thereby enhancing the effect of metformin in inhibiting the proliferation of tumor cells.
