Abstract:
The present application relates to a Yarrowia lipolytica genetically engineered bacterium capable of producing β-elemene and a construction method therefor, and a method for preparing β-elemene. The construction method comprises: integrating the Yarrowia lipolytica endogenous HMGR gene, optimized germacrene A synthase HaGASL101WT412SD433NW436A gene, Yarrowia lipolytica endogenous ERG19 gene, Yarrowia lipolytica endogenous ERG13, and HaGASL101WT412SD433NW436ALERG20 fusion gene of farnesyl diphosphate synthase ERG20 into Yarrowia lipolytica, so as to obtain the Yarrowia lipolytica genetically engineered bacterium capable of producing β-elemene. The method can enable carbon flux to flow into a β-elemene synthesis pathway as much as possible, reducing substrate waste and greatly increasing the yield of β-elemene; and the engineered bacterium has good subculture stability, and can be used for large-scale commercial production.
