Abstract:
The present invention relates to a yeast expression vector comprising: (a) a sequence homologous to the flanking sequence of a target gene fragment; (b) an expression module comprising (i) a multiple cloning site (MCS), (ii) a promoter of Saccharomyces cerevisiae glyceraldehyde-3-phosphate dehydrogenase 3 (ScTDH3), and (iii) a terminator of ScADH1 (alcohol dehydrogenase 1); and (c) a URA3 excision module comprising (i) a promoter of ScURA3, (ii) an open reading frame (ORF) of ScURA3 operably linked to the promoter, and (iii) a terminator of ScURA3 coupled to the promoter and the both ends of the ORF, and a transformation method using the same. The transformation method using the vector of the present invention allows the preparation of a transgenic yeast strain in which a target gene is stably integrated into a chromosome, enables continuous transformation which induces the modification of another target gene in a yeast strain, and can thus be applied usefully to the industrial production of desired products (biomass).
