Abstract:
PURPOSE: To obtain a new [[4-substituted acetyl-o-phenylene]dioxy]diacetic acid derivative useful as a synthetic intermediate for heterocyclic compounds, containing nitrogen and useful for preventing thrombotic diseases, e.g. diseases such as cerebral infarction, cardiac infarction, angina pectoris or peripheral arterial obstruction.
CONSTITUTION: The [[4-aminoacetyl-o-phenylene]dioxy]diacetic acid derivative and [[4-azidoacetyl-o-phenylene]dioxy]diacetic acid derivative of formula I (R is H, a lower alkyl or a phenyl-lower alkyl; R1 is amino or azide) and their salts, e.g. diethyl [[4-aminoacetyl-o-phenylene]dioxy]diacetate and diethyl [[4- azidoacetyl)-o-phenylene]dioxy]diacetate. The compounds of formula I are obtained by reacting a haloacetylcatechol of formula II (Hal is a halogen) with sodium azide of the formula NaN3, providing an azidoacetylcatechol of formula III and then reacting the resultant compound with a (substituted) alkyl haloacetate of formula IV (X is a halogen) in the presence of a base.
