Abstract:
PROBLEM TO BE SOLVED: To provide homogeneous antibody populations.SOLUTION: The present invention is generally directed to methods of producing an increase in the enrichment and/or recovery of preferred forms of monoclonal antibodies. More particularly, the invention relates to methods for eliminating disulfide heterogeneity in the hinge region of recombinant IgG2 antibody proteins. A monoclonal IgG2 antibody is provided, comprising: a light chain polypeptide; and a heavy chain polypeptide having a hinge region, where the antibody comprises an amino acid modification in the heavy or light chain polypeptide such that the modification provides a light chain polypeptide that primarily forms an interchain disulfide bond with amino acids in the hinge region of the heavy chain polypeptide through the most C-terminal cysteine residue of the light chain polypeptide.SELECTED DRAWING: Figure 1B
