To provide single-chain rare-cutting endonucleases, called meganucleases, which recognize and cleave a specific nucleotide sequence, polynucleotide sequences encoding the rare-cutting endonucleases, a vector comprising the polynucleotide sequences, a cell or non-human animal comprising the polynucleotide sequences or rare-cutting endonucleases, and a process for producing the rare-cutting endonucleases.
The single-chain meganuclease capable of causing DNA cleavage comprises first and second domains in the orientation from N-terminal toward C-terminal, wherein the first and second domains are derived from I-CreI mono-dodecapeptide meganuclease, each of the first and second domains comprises a polypeptide fragment consisting of a dodecapeptide motif and a DNA binding moiety, and the first and second domains are linked via a linker.
