Abstract:

PROBLEM TO BE SOLVED: To obtain an isolated DNA encoding FseI restriction endonuclease and capable of producing this enzyme which is one of restriction enzymes and is a tool essential for gene manipulation and is obtained from plasmid pRMFseR 2.
SOLUTION: This new isolated DNA can be obtained from plasmid pRMFseR 2 and encodes FseI restriction endonuclease and is useful for production, or the like, of FseI restriction endonuclease which is one of restriction endonucleases as a tool essential for gene manipulation in modern genetic research. The DNA is obtained by amplifying a gene derived from Frankia- species (NRRL-18528) by using a degenerate primer based on a preserved amino acid sequence of a methylase gene in combination with a degenerate primer based on partial protein sequence data of endonuclease and cloning the endonuclease gene in combination with the methylase gene.
