Abstract:

PURPOSE: To prepare the titled compound useful as a thrombolytic agent, etc., by contacting a liquid containing crude urokinase with an adsorbent composed of L-pyroglutamyl-L-lysyl-L-leucyl-L-argininal bonded to a water-insoluble carrier, and eluting the absorbed substance.

CONSTITUTION: An adsorbent for adsorbing urokinase composed of L-pyroglutamyl-L-lysyl-L-leucyl-L-argininal bonded to a water-insoluble carrier is packed in a column, and made to contact with a liquid containing crude urokinase to effect the adsorption of urokinase to the adsorbent. The adsorbed urokinase is desorbed and eluted with an eluant such as 0.2M citric acid solution, etc. to obtain highly pure urokinase. The insoluble carrier is, e.g. carboxylic acid-type acidic ion exchange resin, carboxymethyl-cellulose, agarose, dextran, etc.
