Abstract:
To produce antipodes of O-demethyltramadol and to provide a method for using an antipode as an analgesic.
Antipodes of 0-demethyltramadol are produced as follows. After a racemic tramadol salt is converted into the base, the (-)-antipode of tramadol is separated by precipitation using L-(+)-tartaric acid, the tramadol base is freed from the precipitant, and subsequently the base is converted into the (-)-antipode of O-demethyltramadol using diisobutylaluminum hydride. From the mother liquid of the tartaric acid precipitation, the (+)-antipode of O- demethyltramadol is produced by freeing the tramadol base and subsequently reacting the base with diisobutylaluminum hydride.
