Abstract:
Provided are a primer composition and an application therefor. The primer composition comprises an upstream primer, a downstream primer, and a sequencing primer, wherein the nucleotide sequence of the upstream primer is as shown in SEQ ID NO.1, the nucleotide sequence of the downstream primer is as shown in SEQ ID NO.2, and the nucleotide sequence of the sequencing primer is as shown in SEQ ID NO.3. By selecting six CpG loci in an SLC30A8 gene promoter specific region, a genomic DNA is treated with hydrosulphite, a treated product serves as a template, amplification is performed on the SLC30A8 gene promoter specific region and the methylation ratio of the six loci is detected by means of pyrosequencing, experimental condition parameters are optimized, the steps and conditions cooperate with each other, and the changes of loci and frequencies are detected accurately and qualitatively.
