Abstract:

PURPOSE: To obtain new compound derivatives containing cyclopropane radicals characterized by a marked intestinal calcium transport activity and exhibiting a lower activity to mobilize calcium from bone, as medicaments for osteoporosis or the like useful for prophylaxis, treatment or the like of bone metabolism disorder.
CONSTITUTION: A compound of formula IV (R1 is H or OH) is obtained by treating ethyl acetoacetate with 1,2-bromoethane in dimethyl sulfoxide in the presence of potassium hydroxide to form cyclopropane ketoester of formula I (Et is ethyl), deriving it to an alcohol by reduction of the ester after protection of the ketone as ketal, converting a hydroxyl radical of the alcohol to phenylsulfide, then, reacting the ketal after deprotection with a Grignard reagent to form tertiary alcohol, oxidating the sulfide after protection of the hydroxyl radical to form sulfone of formula II (Ph is phenyl; TES is triethylsilyl), reacting the resulting compound with aldehyde of formula III (me is methyl) after deprotonation, reducing the resulting compound after acethylation and deprotecting the hydroxyl radical.
