Abstract:
PURPOSE: To obtain a new compd. having selective affinity for the 5-HT receptor of a brain and much smaller affinity for other receptor and useful to treat motion sickness and emesis. CONSTITUTION: This compd. is represented by formula 1, wherein R<1> is H, a 1-4C alkyl, 3-4C alkenyl, cyclopropylmethyl, aryl-1-4C alkyl, (CH2 )n S-1-4C alkyl, CO-R<4> or (CH2 )n -CO-NR<5> R<6> (1<=n<=4, R<4> is H, an alkyl, haloalkyl, alkoxy or phenyl, each of R<5> and R<6> is H, an alkyl or cycloalkyl, but when one of R<5> and R<6> is a cycloalkyl, the other is H), R<2> is H, a 1-4C alkyl, cyclopropylmethyl or 3-4C alkenyl, R<3> is H, a 1-4C alkyl or an amino protective group, HET is (substd.) tetrazolyl or an arom. 5- or 6-membered heterocyclic group having 1-3 hetero atoms (O, N, S). It is, e.g. (-) (4R)-6-(2-oxazolyl)-4-(di-n- propylamino)-1,3,4,5-tetrahydrobenz[cd]indole.
