Abstract:

PROBLEM TO BE SOLVED: To obtain a novel alkali pectase lyase that manifest excellent pectase lyase activity under high alkali conditions, can be produced in an industrial scale and is useful as an enzyme for detergent formulations, fiber treatment enzymes and the like.
SOLUTION: This enzyme is obtained by culturing a micro-organism in Bacillus, e.g. Bacillus sp. KSM-P539 (FERM P-15991) and has the following enzymologic properties: the action: it cleaves the α-1,4-bond in a pectin (e.g. polygalact-uronic acid) through the β-elimination reaction and gives a double bond to the C4-C5 position in the non-reducible terminal to produce an unsaturated oligogalacturonide; the substrate specificity: it acts on (proto)pectin and (acid-soluble) pectic acid; the optimal reaction: the enzymatic reaction is optimally carried out at 30°C at 11 pH (in a 50 mM glycine-sodium hydroxide buffer solution) in the substrate of pectic acid; the stable pH: 4-8; the optimal reaction temperature: at about 45°C; heat resistance: resistant to about 40°C; the molecular weight: about 16 kDa; the isoelectric point: near 10 pH.
