PURPOSE: To provide a novel aryl-substd. alkoxycarbonyl-1,4-dihydropyridine-5- carboxylic acid ester being a calcium route ligand having the selectivity related to the L-type calcium route of the central nervous system and useful for treating central nervous diseases.
CONSTITUTION: A compd. represented by formula I (wherein R1 is a 1-8C alkyl, benzyl or 3-8C cycloalkyl; R2 is a halogen, trifluoromethyl or cyano; R3 is R2 or H; and b are each 0-4; R4 and R5 are each H, a 1-6C alkyl or phenyl; A is O, -CO-O-, -O-CO-, -NH-CO or -CO; and R6 is a heterocyclic group, benzyl or a 6-10C aryl) and its salt, for example, methyl 5-[2-(benzoyloxy)- ethoxycarbonyl]-2,6-dimethyl-4-(2,3-dichlorophenyl)-1,4-dihydropyridine-3- carboxylate represented by formula II are designated. The compd. represented by the formula I is obtained, for example, by reacting a compd. represented by formula III with a compd. represented by formula IV (wherein B is O, -CO- or the like) in an inert solvent in the presence of a base.
