PURPOSE: To obtain a danoxamine which is nontoxic even in high concentrations and can be satisfactorily used as a drug against iron accumulation diseases, Alzheimer's disease, etc., on an industrial scale by treating a salmycin obtained by fermenting microorganisms.
CONSTITUTION: Streptomyces violaceus 37290 (DSM 8286) are fermented to obtain salmycins A to D represented by formula I. Salmycins A or B is hydrolyzed to prepare a danoxamine of formula II. Salmycins A to D are subjected to hydrolysis and a treatment with a metal scavenger or an ion binding agent or treated by reduction with Fe3 ions to prepare a nordanoxamine of formula III. The hydrolysis is performed under gentle conditions, desirably at a pH of 9.5, at room temperature in, for example, ammonium hydroxide. The danoxamine and the nordanoxamine can be separated and purified by e.g. chromatography.
