PURPOSE: To provide a D-amino acid oxidase having broad substrate specificity and useful as a clinical diagnostic agent for renal insufficiency.
CONSTITUTION: This D-amino acid oxidase is produced by the cultivation of a microorganism belonging to the incomplete fungi Curvularia, e.g. NF02201 strain (FERM P-14017) and has the following properties. (1) It oxidizes D-amino acids to produce hydrogen peroxide, 2-oxo acids and ammonia, is strongly reactive with D-alanine, D-valine, D-phenylalanine, D-methionine, D-isoleucine, D-leucine and D-glutamine and is reactive with D-serine, Dasparagine, D- threonine and D-glutamic acid, (2) it has an optimum pH of 9-10 and is stable in the pH range of 7-10 by the treatment at 30°C for 30min; (3) it has an optimum temperature of about 35°C and is stable up to 35°C by the treatment at pH8.5 for 10min; (4) the molecular weight is about 150,000 (gel-filtration) and about 39,000 (by SDS-polyacrylamide electrophoresis); (5) the isoelectric point is 4.9.
