Abstract:

PROBLEM TO BE SOLVED: To obtain the subject new DNA encoding a recombinant protein having a sequence of adding a basic amino acid to the N-terminal region of a wild type chicken lysozyme signal peptide having a specific amino acid sequence and capable of being produced in a host cell such as of a yeast, in a large amount.
SOLUTION: This DNA is a new DNA encoding a modified signal peptide consisting of an amino acid sequence obtained by adding at least one basic amino acid to the N-terminal region of the amino acid sequence of a wild type chicken lysozyme signal peptide, can widely increase the amount of a recombinant type protein in a host cell such as of a yeast, and can realize a cost down, or the like, of various recombinant type protein preparations such as a cytokine. The DNA is obtained by preparing a primer used for modifying the gene based on the amino acid sequence of the objective modified signal peptide by a phosphate diester method, or the like, and amplifying the DNA encoding the wild type chicken lysozyme signal peptide by a PCR method by using the obtained primer.
