PURPOSE: To prepare a diagnosticum of periodontal diseases by specifically hybridizing DNA of Actinobacillus actinomycetemcomitans (A. a. bacterium) to produce DNA probe capable of discriminating serum type of bacteria.
CONSTITUTION: An antigen protein gene of cell of Actinobacillus actinomycetemcomitans is extracted and collected from one kind of serum type bacterium belonging to the genus Actinobacillus actinomycetemcomitans and λL47 phage, etc., is used as a vector to prepare genomic library. Then the genomic library is screened by using serum of ADP patient as a primary antibody by immunochemical method to give a clone showing positive reaction to the serum of ADP patient from phage plaque. The clone is used as DNA probe. The probe is specifically hybridized with DNA of bacterium of Actinobacillus actinomycetemcomitans to discriminate each serum type of bacterium of Actinobacillus actinomycetemcomitans.
