To obtain the subject polypeptide identified as a new hRAD 54 by homology with yeast RAD 54 protein and useful for treating xeroderma pigmentosum and Bloom's syndrome, and cancer-prone syndromes such as X- linked Mental Retardation with-thalassemia Syndrome and Vernet's syndrome, and for treating cancers such as breast cancer.
This hRAD 54 is a polypeptide having an amino acid sequence of the formula, and belongs to the SNF2 superfamily of DNA and RNA helicase. The hRAD 54 is obtained by isolating the code region of a hRAD 54 gene with an oligonucleotide screened on the basis of a known DNA sequence, screening a human cDNA library, etc., with a labeled oligonucleotide having a sequence complementary to the sequence of the hRAD 54 gene, and subsequently subjecting the obtained polynucleotide to a DNA recombination technology.
