PURPOSE: To detect the anti-LSP antibody in the serum of liver disease patients and make it helpful for treatment by measuring anti-LSP antibody by an enzyme immunity method with the hepatocellular membrane conjugated protein (LSP) extracted from animals other than man as an antigen.
CONSTITUTION: LSP is collected from the fresh liever of, for example, a pig, and it is concd. to 5mg/ml protein concn. On the other hand, the enzyme mark LSP antibody attaching peroxidase to antihuman LgG-goat IgG is prepd. Said pig LSP soln. is fixed to polystyrene beads to make LSP beads. The enzyme labelling antihuman IgG-goat IgG and the human serum to be examined are added into a test tube contg. one piece of said beads, after whih a buffer contg. H2O2 is added to allow reaction to take place. After prescribed time of reaction, a reaction stopping agent is added and the absorbancy of 418mm wavelength is measured. The amount of the anti-LSP antibody is known by using the calibraction work obtd. by using the serum contg. known LSP antibody.
