Abstract:

PURPOSE: To readily obtain the subject cyanohydrins in a relatively high optical purity and yield by adding hydrogen cyanide to a carbonyl compound in the presence of hydroxynitrile lyase, under a specific condition.
CONSTITUTION: A homogeneous aqueous solution of hydroxynitrile lyase is buffered with an acetate buffer having a buffer concentration of 0.005-0.1 mol/L or with a non-acetate buffer. An adding reaction of hydrogen cyanide to a carbonyl compound (aldehydes or ketones) is performed in a two-phase solvent system in which a volume ratio of an organic solvent at least substantially non-miscible with water to the homogeneous aqueous solution is about 5/1-about 1/5. A solution of hydrogen cyanide and a solution of the carbonyl compound in an organic solvent are properly mixed during the reaction period with the homogeneous aqueous solution of hydroxynitrile lyase.
