PURPOSE: To provide a method for simply detecting hepatitis C virus (HCV) with a high sensitivity and reliability for which a polymerase chain reaction (PCR) technique is utilized.
CONSTITUTION: This method for simply detecting hepatitis C virus is to carry out steps from an extraction step for HCV from a specimen to the completion of a PCR at the second time in only one reactional tube. In the method, a sense primer having a base sequence of 5'-ACTCCACCATAGATCACTCC-3' and an antisense primer having a base sequence of 5'- AACACTACTCGGCTAGCAGT-3' are used in the PCR at the first time and a sense primer having a base sequence of 5'- TCTTCACGCAGAAAGCGTCTAGCCA-3' in a 5'-nontranslation region and an antisense primer having a base sequence of 5'- CAATTCCGGTGTACTCACCGGTTCC-3' are used in the PCR at the second time to respectively set the final concentration of Mg2+ at 6.2 and 4.5mM in the PCRs at the first and the second times and the final concentration of the KCl at 120-140mM in the PCR at the first time.
