Abstract:
Quantitative measurement of analytes in a blood plasma sample by immunochromatography, in which the sample is deposited in the presence of the control reagent and conjugate or conjugate mixture, is new. A capture reagent: is immobilized on the support, forms a bond pair with the analyte at capture zone (2) level, and forms a bond with control reagent carrying a detectable marker at control field of ratiometric measurement zone (7). The signal is produced by a fixed conjugate in the capture zone level and a control signal is produced by the control reagent at the ratiometric measurement zone level. The signal is produced by a fixed conjugate in the capture zone level and a control signal is produced by the control reagent at the ratiometric measurement zone level. The quantity of the analyte in the sample is directly proportional to the ratio of the test signal. The conjugate or conjugate mixtures or the control reagent are used in dry form. The control reagent is immunoglobulins of animal origin and the capture reagent is an antigen/antibody directed against the animal species of the control reagent. The marker used on the conjugate or conjugate mixture and on the control reagent are identical colloidal gold particles. The reading of the test signals and the ratiometric reflectance/transmittance measurement are carried out with a reader strips and compared to a calibration curve established from the analyte with several concentrations. The calibration curve is directly proportional to the ratio analyte signal/ratiometric signal. The passage of the markers is automatically detected in zone of the strip to the beginning of the migration of the sample. A period of time (10 minutes) is detected from the detection of the passage. The signals generated by the markers having an absorption line and control of the strip is measured by automatic acquisition and collected by photodetectors. Independent claims are also included for: (1) a kit for implementation of quantitative measurement of an analyte; and (2) a device for the execution of quantitative measurement of an analyte.
