Abstract:

PROBLEM TO BE SOLVED: To obtain a new optically active ceramide bearing an optically active hydroxyl group on the 3-site of the fatty acid segment by reaction between an optically active specific diol and an optically active specific ester in the presence of a basic catalyst while distilling by-product lower alcohol off the reaction system.
SOLUTION: This optically active compound is expressed by formula I (R1 is a 9-19C saturated aliphatic hydrocarbon; * denotes an asymmetric carbon atom with S- or R-configuration), e.g. (2,3R,3'R)-2-(3'-hydroxyoctadecanoyl) aminooctadecane-1,3-diol. This compound of formula I is obtained by the following process: a diol of formula II is dissolved in 1 to 10wt. times of an alcohol (e.g. ethanol), and an ester of formula III is dripped at 1.0-1.5 mol per mol of the diol of formula II into the resultant solution in the presence of a basic catalyst such as sodium hydroxide at 50-120°C under a pressure of 30-100mmHg to carry out a reaction while expelling byproduct lower alcohol out of the system to effect deposition of crude crystal which is then recrystallized and purified.
