Abstract:
PURPOSE: To efficiently prepare the subject compound useful as a synthetic intermediate or the like for a calcium channel inhibitor diltiazem in good yield by acylating a racemic compound of a specific phenylpropionic acid derivative with a lipase as a catalyst in an anhydrous organic solvent.
CONSTITUTION: A racemic compound of a phenylpropionic acid derivative represented by formula I [X is NO2, NH2, NHCOCH3, NHCOCF, NHCO2CH3, NHCO2C(CH3), or NHCHO; R1 is an alkyl] is acylated with a lipase derived from Pseudomonas cepacia as a catalyst in an anhydrous organic solvent to thereby produce a (2S,3S)-threo-alkyl-2-hydroxy-3-(4-methoxyphenyl)-3-(2-X- phenylthio)propionate represented by formula II and a (2R,3R)-threo-alkyl-2- acyloxy-3-(4-methoxyphenyl)-3-(2-X-phenylthio)propionate represented by formula III.
