Abstract:
PROBLEM TO BE SOLVED: To provide a method for readily producing an antitumor platinum complex of carboplatin useful as an antitumor agent, etc., in a high yield by reacting cis-dichlorodiammine platinum complex with 1,1-cyclobutanedicarboxylic acid (salt) solution prepared by neutralization in an aqueous solution under heating.
SOLUTION: The objective antitumor platinum complex of carboplatin of the formula is readily obtained in high yield by reacting cis-dichlorodiammine platinum complex with 1,1-cyclobutanedicarboxylic acid or its salt prepared by neutralizing the one to pH7.0-7.5 so that the using amount of the 1,1- cyclobutanedicarboxylic acid or its salt may be 1-4mol based on 1mol cis- dichlorodiammine platinum complex by using water as a solvent at the reaction temperature within the range of 70 to 100°C to provide the carboplatin, suspending the product of carboplatin in N,N-dimethylformamide, and stirring the suspension of the carboplatin at 20-50°C to purify the carboplatin. The obtained carboplatin includes a silver salt of under the limit of detection by flameless atomic absorption analysis and ≤0.1wt.% content of the cis- dichlorodiammine platinum measured by high-performance liquid chromatography.
