PURPOSE: To obtain the subject compound for medical use, having vasodilator action and protecting action on heart muscle by reacting an o-(aminoalkylamino) thiophenol with a specific glycidic acid ester in the presence of an iron ion and subjecting the reaction product to ring closure reaction.
CONSTITUTION: An o-(aminoalkylamino)thiophenol compound of formula I [Y is halogen, alkyl, alkoxy, aryl or aryloxy; (n) is 1-6; R1 is amino group of the formula NR1aR1b (R1a and R1b are H, 1-6C alkyl, aryl or R1a is linked to R1b together with nitrogen to be bonded to them to form cyclic amino group or aromatic heterocyclic group)] is reacted with a trans-3-(4-methoxyphenyl)glycidic acid ester of formula II (R2 is lower alkyl) in the presence of a ferrous or ferric ion in a nonpolar solvent under heating to produce a threo derivative of a compound of formula III stereoscopically selectively, the ester group is hydrolyzed and the resulting substance is subjected to ring closure reaction to give the objective benzothiazepine derivative of formula IV in a high yield.
