To obtain a new DNA molecule for e.g. efficiently producing a nucleoside analogue synthase with few subreactions, comprising a purine nucleoside phosphorylase gene derived from Bacillus thermophilic bacteria and encoding a specific amino acid sequence.
This new DNA molecule is derived from Bacillus thermophilic bacteria and contains a purine nucleoside phosphorylase structural gene encoding an amino acid sequence of the formula. This new DNA is useful for e.g. the mass production, through recombinant DNA technique, of nucleoside phosphorylase enabling efficient nucleoside analogue synthesis with few subreactions. This DNA molecule is obtained by the following process: a gene encoding nucleoside phosphorylase derived from Bacillus thermophilic bacteria [e.g. Bacillus stearothermophilus TH 6-2 strain (FERM P-2758)] is sectioned by a plasmid followed by subcloning and then binding to a vector to prepare various clones, which are then screened.
