PURPOSE: To industrially and advantageously purify a high-purity albumin from a blood collected at large amounts by lowering ion strength of supernatant prepared by pre-treating plasma to control application condition and fractionating the supernatant by a method using an anion chromatography.
CONSTITUTION: A frozen plasma 1 is defrosted (a) and centrifuged (b) to remove precipitate and a supernatant 2 is collected and diluted with water to lower ion strength and control application (c) condition and then subjected to anion column chromatography 4 and hemoglobin is subjected to passing-through fractionation 5 in condition of ion strength of 0.01-0.03 (e) and then transferrin 6 is eluted in condition of ion strength of 0.04-0.08. Further, the ion strength is raised to 1.00 to elute ceruloplasmin 8 and then ion strength is lowered to elute albumin fractionation by non-equilibration of Gelbet and the albumin fractionation is subjected to pH treatment 9 and simultaneously cleaned by an auxiliary filtration 10 using Celite (i) and then desalted by ultrafiltration 13 to provide the albumin 20 purified in high purity.
