Abstract:

PROBLEM TO BE SOLVED: To provide a method for searching a natural peptide forming amyloid fiber by a template reaction with a small amount of the nucleus of a fibrillated amyloid β-protein to form amyloid fiber and amplifying the fiber to increase its volume, and designing and developing a new artificial peptide usable as a substitute for the natural peptide, a method for the amplification of amyloid fibrosis of the amyloid β-protein, a reagent for the amplification reaction, a method for detecting diseases caused by amyloidosis and a reagent for the detection method.
SOLUTION: The amplification of amyloid fibrosis of amyloid β-protein is carried out by using a reagent containing a peptide composed of the 14th residue to the 23rd residue of amyloid β-peptide or a peptide obtained by substituting all positive-charge side chain amino acids of the above peptide with Lys and all negative-charge side chain amino acids with Glu. The invention further provides a reagent for the amplification method, a method for the detection of diseases caused by amyloidosis using the reagent containing the peptide, a reagent for the detection method and a new artificial peptide usable in the above reagents.
