PURPOSE: To obtain a red dye-containing callus which is rich in the dye and gives it inexpensively, by introducing the callus through tissue culture of Raphanus sativus, L., selecting the red calluses excellent in red dye-producing capability, culturing and proliferating them.
CONSTITUTION: The tissue of Raphanus sativus, L. such as seed or leaf is cultured in the MURASHIGE-SKOOG medium to induce calluses. Then, red calluses excellent in red dye production capability are selected from them and the selected calluses are cultured and proliferated to give red dye-containing calluses. The calluses can be directly used as a natural dyeing agent for fibers. When the dye is required to be purified, the calluses are ground, passed through ODS columns to absorb the red dye and the pigment is eluted with aqueous methanol whereby the dye is isolated.
