SEPARATION AND PURIFICATION OF ASTAXANTIN FROM CULTURED FUNGAL CELL

Abstract:

PURPOSE: To industrially and advantageously separate and purify astaxantin useful for foods or medicines, etc., by treating cultured fungal cell of Phaffia rhodozyma combinedly using a supper critical carbon dioxide extracting liquid in which ethanol is used as an assistant solvent and a silver nitrate solution.

CONSTITUTION: This astaxantin is industrially and advantageously separated and purified from a cultured fungal cell of Phaffia rhodozyma by culturing Phaffia rhodozyma ATCC24202 strain in a culture medium at 20°C for 96hr using a rotary shaker and harvesting the fungus by centrifugation, lyophilizing the fungal cell, adding acetone thereto, extracting the fungal cell while crushing the cell with a mortar, cooling the extracted liquid to -50°C, filtering the resultant precipitate, adding silver nitrate solution to the filtrate, further, adding ethanol which is an assistant solvent thereto, putting these liquids into an equilibrated cell 10 of supper critical extraction apparatus, feeding carbon dioxide from a gas cylinder 1 by the operation of a valve to completely replace the interior of the system with carbon dioxide, raising the pressure in the system using a pump 5 for increasing pressure, charging the supercritical fluid under heating by a heating unit 13 and recovering the extract into a trap 14.
